    Control system and automation of the spectrum acquisition for a perturbed gamma-gamma angular correlation spectrometer

    This work reports results from the automation of data acquisition for a gamma-gamma perturbed angular correlation (PAC) spectrometer located in the hyperfine interactions laboratory (LIH) of IPEN. A PAC spectrometer comprises usually four scintillation detectors (BaF3 for example) and carries out time measurements from gamma rays of any two combinations of detectors in coincidence. Gamma rays come from the decay of probe nuclei embedded into the material to be studied. Measurements are generally performed as a function of temperature. The fourth-detectors PAC spectrometer was automated in Java language because it is a multiplatform and object oriented programming language, having, therefore, portability and longer lifetime due to the ease of implementation of peripherals. Finally, the software has a friendly and intuitive interface with the user being able to operate the hardware manually or automatically with time acquisitions up to 100 consecutive hours. Before each acquisition, the software sets the temperature, waits for temperature stability and records data in different files. Moreover, the software also saves in a file all information of relevant parameters during acquisition.

    Investigation of hyperfine interactions in immunoglobulin G with perturbed angular correlation spectroscopy using 111Cd

    Hyperfine interaction parameters have been used to investigate physical properties of biomolecules from different isotypes of immunoglobulins G (IgG1, IgG2a and IgG2b). Immunoglobulin IgG1 induces a pattern of response type Th2 that corresponds to an allergic immunological response, whereas immunoglobulin IgG2 induces a pattern of response Th1 that corresponds to a cellular immunological response, suited for response against parasites. Hyperfine interactions were measured by perturbed gamma-gamma angular correlation (PAC) spectroscopy using 111In(111Cd) and 111mCd(111Cd) as probe nuclei. The measurements were carried out for both the complete and fragmented molecules at 295 K and 77 K, and both static as well as dynamic interactions were observed. Results of both measurements were used to calculate the rotational correlation time for each studied molecule. The complete and fragmented molecules showed different dynamic interactions. It was therefore conclude that the PAC technique is quite adequate for differentiating between the isotypes of immunoglobulins as well as their dynamic behavior if adequate nuclear probe is utilized with rigorous control of measuring conditions. The hyperfine parameters obtained are related to the physical properties of each molecule in an attempt to understand the difference in their biological function.

    A doen¸ca de Chagas ´e uma parasitose tecidual e hem´atica, tem evolu¸c˜ao crˆonica e ´e causada pelo protozo´ario Trypanosoma cruzi que ´e transmitido ao homem e a v´arios mam´ıferos sens´ıveis por hem´ıpteros hemat´ofagos da subfam´ılia Triatominae. Esta tripanossom´ıase est´a entre os principais problemas de sa´ude da Am´erica do Sul, afetando milh˜oes de pessoas. Devido `a ausˆencia de vacinas ou disponibilidade de tratamento espec´ıfico, diferentes linhas de estudos s˜ao tra¸cadas por pesquisadores para sanar este problema. Em nosso estudo optamos pelo emprego da t´ecnica de PAC, pois esta caracteriza intera¸c˜oes hiperfinas dos campos el´etricos e magn´eticos das biomol´eculas com os momentos el´etrico e magn´etico do n´ucleo de prova, fornecendo dessa forma, conhecimentos a respeito do comportamento das biomol´eculas e a melhor compreens˜ao de fenˆomenos biol´ogicos relacionados com a dinˆamica metab´olica dos sistemas biol´ogicos. Tendo em vista esta possibilidade utilizamos como materiais biol´ogicos os DNAs e anticorpos de diferentes linhagens de camundongos infectados pela cepa Y de T. cruzi e como n´ucleo de prova o 111Cd. Os resultados obtidos, com amostras dilu´ıdas em ´agua e a temperatura ambiente, demonstraram existir uma intera¸c˜ao dinˆamica entre o material biol´ogico e o n´ucleo de prova, sendo que esta intera¸c˜ao relaciona-se com o parˆametro λ, que provoca o decrescimento exponencial da fun¸c˜ao de perturba¸c˜ao. Estes resultados contribuir˜ao grandemente para a explica¸c˜ao dos diferentes padr˜oes de resposta imunol´ogica apresentados pelas diferentes linhagens de camundongos quando infectados pela cepa Y de T. cruzi.
