    Aplicações na área da saúde e meio ambiente desenvolvidas no LEER

    O Laboratório de Espectroscopia e Espectrometria das Radiações (LEER) realiza pesquisas em Física Aplicada na área da saúde, nutrição, meio ambiente e correlatas utilizando técnicas nucleares e atômicas. As investigações são realizadas utilizando as técnicas de Espectroscopia e Espectrometria de raios gama, Ativação Neutrônica e Fluorescência de Raios-X. O LEER atua também na área de Ensino em Física, na implementação de métodos didáticos e na divulgação cientifica da física das radiações. Estas atividades envolvem estudantes de todos os níveis, de Iniciação Científica ao Doutorado, bem como Pós – doutorando e Cooperações de âmbito Nacional e Internacional com Instituições de referência (IF-USP, IBu, Unisa, UFF, Unifesp, HU-USP, Unicid, Unicamp, UNSL/San Luis-Argentina e Udelar/Montevideo-Uruguay). O programa de atividades estabelecido no LEER caracteriza-se pela interação entre as diferentes áreas da ciência que atuam no âmbito da saúde e meio ambiente, incluindo: Elaboração de propostas tecnológicas para a melhoria da qualidade vida de populações carentes e desprovidas de atendimento médico; Caracterização multielementar de novos medicamentos, vacinas e soros (antivenenos); Elaboração de propostas que possam suprimir/minimizar ou agregar melhorias no uso de modelos animais; Estudo das alterações ecológicas e os processos ambientalmente danosos relacionando-os ao ecossistema que incorporem as dimensões físicas, químicas e toxicológicas. Esses temas compõem monografias, dissertações e teses contribuindo para formação recursos.

    Determination of inorganic elements in blood of mice immunized with Bothrops Snake venom using XRF and NAA

    In this work, mice genetically modified [HIII line] were immunized against different Bothrops snake venoms to produce anti-Bothrops serum (antivenom). The Neutron Activation Analysis (NAA) and Energy Dispersive X-Ray Fluorescence (EDXRF) techniques were used to evaluate Ca and Fe concentrations in blood of these immunized mice in order to establish a potential correlation between both phenotypes: antibody response and blood constituents after Bothrops venom administration. The results were compared with the control group (mice not immunized) and with human being estimative. These data are important for clinical screening of patients submitted to immunological therapy as well as the understanding of the envenoming mechanisms.

    Ions concentration in blood samples of SJL/J dystrophic mice strains using X-ray fluorescence spectrometry

    Star This study proposes an investigation of ions in whole blood of the dystrophic animal model SJL/J (mice strain with dysferlin protein deficiency) and in the control group (C57BL/6J) using the Energy Dispersive X-ray Fluorescence (EDXRF) spectrometry technique. The comparison between control and dystrophic mice results shown an increase in blood for P, S, K and Fe (p < 0.05) while a decrease in Ca (p < 0.05). This elemental analysis will contribute to evaluate the best diagnostic, care and treatment procedures, for the Progressive Muscular Dystrophy.

    Uso de mini-espectrômetro de fluorescência de raios-x como alternativa para prática clínica de dialisados

    O objetivo do presente estudo foi avaliar a performance da técnica de Fluorescência de Raios-X por Dispersão de Energia (FRX-DE) para análise de íons, de relevância clínica (Ca, Cl, K, Fe), em sangue total de pacientes com insuficiência renal crônica (IRC) submetidos a tratamento dialítico. Com os dados do presente foi possível elaborar uma discussão sobre as vantagens e limitações do uso deste procedimento para a realização desses exames bioquímicos em Centros de Hemólise. Durante a investigação as concentrações obtidas para Ca, Cl, K e Fe levaram a resultados que corroboram com o quadro clínico obtido pelas análises convencionais.

    Correlation analysis of inorganic elements in whole blood of GRMD dogs using INAA

    Instrumental neutron activation analysis technique (INAA) has been used to determine Br, Ca, Cl, Cr, Fe, K, Mg, Na, P and S concentrations in whole blood of GRMD dogs’ samples from control, carrier and affected dogs (treated and untreated). The GRMD dogs are relevant as an experimental model for Duchenne Muscular Dystrophy (DMD), which is the most severe and prevalent type of muscular dystrophy. Muscle weakness, premature death and instability of the membrane that involves the muscle fibers - causing functional/structural abnormalities and cell death - are main characteristics of this genetic disease. To show in more details the alterations that this disease may cause in whole blood, correlations matrixes were generated for control, carried and affected groups permitting a comparison between them. The results emphasize physiologic differences for K and Mg and suggest that Br results are altered, emphasizing a constant monitoring need. Other than that, these results may help the researchers to evaluate the efficiency of new treatments and to compare the advantages of different treatment approaches before performing tests in patients with muscular dystrophy.

    Progress and trends for radiation applications in clinical practice

    In this study, we intend to optimize the use of equipment based on the X-Ray Fluorescence technology to perform ions dosage in body fluids. This procedure can be an efficient alternative for clinical diagnosis, mainly in underserved regions of Brazil with the deficit of medical care in the hospitals. In addition, this procedure has potential use when the biological material is scarce, case of the pediatric practice in newborns and premature infants (blood collection is the main cause of transfusions) as well as in Hemodialysis units which requires several biochemical tests (before and after dialysis).

    Evaluation of ions and metals in the blood of GRMD dogs submitted to hASCs therapy by NAA and XRF techniques

    The elements Br, Ca, Cl, Cr, Fe, K, Mg, Na, P, Rb, S, and Zn were investigated in the whole blood samples of Golden Retriever dogs submitted to cell therapy (hASCs). These analyses were performed over 2 years using Neutron Activation Analysis and X-Ray Fluorescence techniques. The results were compared with control and untreated dog's. A significant increase was observed in K blood levels. There was also variation in blood levels of Br, Cr, Fe, Rb, S, and Zn.

    Differences in iron concentrations in whole blood of animals model using NAA

    In this study Neutron Activation Analysis technique (NAA) was applied to determine Fe concentrations in whole blood samples of several animals model: mice (Mus musculus), Golden Hamsters (Mesocricetus auratus), Wistar rats, Albinic Rabbits of New Zealand, Golden Retriever dogs and Crioula breed horses. In the health area these animals model are used for several investigations, mainly those that involve testing of new medicines, vaccines, antibiotics, anesthetics, antidepressants, organs transplantation as well as medical diagnostic studies. While small-sized animals model (mice, hamsters, rats and rabbits ) are very convenient to perform medical investigation due to the low cost, easy handling and medico-legal implications, medium and large sized animals, (mainly Golden Retrievers dogs) are selected for medical diagnostic studies because of their physiological similarities with the humans. Particularly, in this study, horses (Crioula breed) were also investigated because they are frequently used for antivenom production in Brazil. An important aspect to be considered during these medical investigations is the needs to check for similarities between the animal’s blood and the human’s blood. Besides, considering that Fe plays important functions in blood (acts as indicator of a great number of anomalies) consulting these data is possible to select the convenient animal model for experiments which the similarities with the humans are an important condition. The NAA measurements were performed in the nuclear reactor IEA-R1 (3.5-4.5MW, pool type) at IPEN/CNEN-SP (Brazil). Fe concentrations results in the blood of these animals were compared with human blood estimative and some significant differences were identified.

    Dosagem de íons em tecido biológicos de camundongos distróficos

    Nesta obra a investigação multielementar de tecidos biológicos (sangue total, ossos e órgãos) de camundongos distróficos, usados como modelo animal da Distrofia Muscular de Duchenne (DMD), foi realizada utilizando a técnica de Análise por Ativação com Nêutrons (AAN). O objetivo teve como foco determinar a concentração de íons de relevância clínica e nutricional (Br, Ca, Cl, K, Mg, Na e S) de camundongos da linhagem distrófica Dmdmdx/J e grupo controle C57BL/6J. Amostras de sangue total, ossos, coração e músculo foram irradiadas no reator nuclear IEA-R1 no IPEN (São Paulo, Brasil). Os resultados constituem as primeiras estimativas para os valores de referência nesses tecidos biológicos e viabilizam a monitoração de diversas funções do organismo saudável (C57BL/6J) e distrófico (Dmdmdx/J). Este livro apresenta dados que poderão auxiliar pesquisadores avaliar e comparar as vantagens e desvantagens dos diferentes tratamentos, realizados na distrofia muscular, quando este modelo animal for empregado, auxiliando na avaliação da eficácia de novos procedimentos terapêuticos antes de serem empregados em pacientes com DMD.
